Abstract(s)
Ribonucleases are fundamental for many cellular and developmental processes and defects in their expression are associated with human disease. XRN1 is a highly conserved cytoplasmic exoribonuclease which degrades RNAs in a 5’-3’ direction and mutations in this gene are associated with osteosarcoma (Zhang et al., 2002).
Our work intends to elucidate novel functional networks through which XRN1 modulates the human transcriptome and assess its potential role in colorectal tumorigenesis. With this purpose, we are studying the effect of XRN1 knockdown on reporters mRNA levels in HeLa and colorectal cancer cell lines. Furthermore, we aim to perform an extensive characterization of the mRNA targets affected by these exoribonucleases knockdown coupled with RNA-seq.
